The article presents materials on improving biotechnological methods of freezing and thawing sperm of rams - producers of the Ordabasy breed of meat and fat sheep. As a result of the studies, high efficiency of insemination of ewes with thawed sperm, frozen and stored in disposable polypropylene (plastic) straws was found out. There is no doubt that the quality of sperm depends on the volume and form of packaging. 0.25 ml straws provide better preservation of sperm during freezing. During freezing and thawing, some sperm are damaged. This has a direct impact on the fertilizing ability of spermatozoa. The results of our studies show that when defrosting from a small container, the survival rate of sperm is higher. This indicator is affected, first of all, by the diameter of the container. In smaller volume straws of 0.25 ml, compared to 0.5 ml packaging, the freezing and subsequent thawing rate is more uniform.
It has been established that this method of freezing the sperm of stud rams provides greater fertility. When inseminating using sperm from disposable polypropylene (plastic) straws of 0.25 and 0.5 ml, before granules, the percentage of fertile inseminations is higher by 8-11% at P < 0.001. This advantage is a direct consequence of the above. The proposed method of freezing and thawing, in which a larger volume of viable spermatozoa is preserved, works better. This is confirmed by the data of our research and the experience of various farms. Our improved method of organizing the continuity of the biotechnological process of collecting, packaging and freezing the sperm of stud rams makes it possible to increase labor productivity, reduce the colonization of sperm by microorganisms, and reduce the costs of artificial insemination of animals without reducing its effectiveness.
